    Investigation of a drag reduction on a circular cylinder in rotary oscillation

    Drag reduction in two-dimensional flow over a circular cylinder, achieved using rotary oscillation, was investigated with computational simulations. In the experiments of Tokumaru & Dimotakis (1991), this mechanism was observed to yield up to 80% drag reduction at Re = 15 000 for certain ranges of frequency and amplitude of sinusoidal rotary oscillation. Simulations with a high-resolution viscous vortex method were carried out over a range of Reynolds numbers (150–15 000) to explore the effects of oscillatory rotational forcing. Significant drag reduction was observed for a rotational forcing which had been very effective in the experiments. The impact of the forcing is strongly Reynolds number dependent. The cylinder oscillation appears to trigger a distinctive shedding pattern which is related to the Reynolds number dependence of the drag reduction. It appears that the source of this unusual shedding pattern and associated drag reduction is vortex dynamics in the boundary layer initiated by the oscillatory cylinder rotation. The practical efficiency of the drag reduction procedure is also discussed

    Evolution and diversity of secretome genes in the apicomplexan parasite Theileria annulata

    <b>BACKGROUND</b>: Little is known about how apicomplexan parasites have evolved to infect different host species and cell types. Theileria annulata and Theileria parva invade and transform bovine leukocytes but each species favours a different host cell lineage. Parasite-encoded proteins secreted from the intracellular macroschizont stage within the leukocyte represent a critical interface between host and pathogen systems. Genome sequencing has revealed that several Theileria-specific gene families encoding secreted proteins are positively selected at the inter-species level, indicating diversification between the species. We extend this analysis to the intra-species level, focusing on allelic diversity of two major secretome families. These families represent a well-characterised group of genes implicated in control of the host cell phenotype and a gene family of unknown function. To gain further insight into their evolution and function, this study investigates whether representative genes of these two families are diversifying or constrained within the T. annulata population. <b>RESULTS</b>: Strong evidence is provided that the sub-telomerically encoded SVSP family and the host-nucleus targeted TashAT family have evolved under contrasting pressures within natural T. annulata populations. SVSP genes were found to possess atypical codon usage and be evolving neutrally, with high levels of nucleotide substitutions and multiple indels. No evidence of geographical sub-structuring of allelic sequences was found. In contrast, TashAT family genes, implicated in control of host cell gene expression, are strongly conserved at the protein level and geographically sub-structured allelic sequences were identified among Tunisian and Turkish isolates. Although different copy numbers of DNA binding motifs were identified in alleles of TashAT proteins, motif periodicity was strongly maintained, implying conserved functional activity of these sites. <b>CONCLUSIONS</b>: This analysis provides evidence that two distinct secretome genes families have evolved under contrasting selective pressures. The data supports current hypotheses regarding the biological role of TashAT family proteins in the management of host cell phenotype that may have evolved to allow adaptation of T. annulata to a specific host cell lineage. We provide new evidence of extensive allelic diversity in representative members of the enigmatic SVSP gene family, which supports a putative role for the encoded products in subversion of the host immune response

    In vitro effect of canine hyperimmune sera on TNFa activity

    [Introduction]: Septic shock in dogs is caused by cardiovascular and vasomotor failure associated with an uncontrolled intrinsic release of inflammatory mediators [1–5]. The syndrome is characterized by cardiovascular dysfunction, vascular permeability alterations, pulmonary oedema and tissue hypoxia resulting from microthrombi which may culminate in disseminated intravascular coagulation and catastrophic multiple organ failure [6,7]. Systemic bacterial infection, particularly by Gram-negative enterobacteria, haemorrhagic trauma, gastric dilation/volvulus and pancreatitis are the major underlying causes leading tosepsis [8,9]. Because of haemodynamic instability and associated hypovolemia, fluid replacement therapy is generally applied to restore effective circulating volume. The use of fresh frozen plasma has been recommended in cases of coagulopathies as it has been recognized to assist restoration of haemodynamic stability [1,5,10,11]. There is increasing evidence that the drivers of the haemodynamic instability are inflammatory mediators (particularly TNFa) activated primarily by bacterial endotoxin [3,4,12,13]

    The effect of equine hyperimmune sera on TNF alpha activity in a L929 cell bioassay

    In this pilot study we examined the effect of sera from a proprietary hyperimmune plasma product (Equiplas) on the activity of TNF: in an in vitro L929 cell bioassay. In brief, we report observations from 2 accessions of sera. Accession 1 describes the antiTNF: activity of 3 hyperimmune sera and an untreated serum sample that were provided blind to the study. Accession 2 reports a comparison of antiTNFalpha activity found in 3 paired hyperimmune sera collected following a multiple endotoxin vaccination regimen

    Pre-transplant CDKN2A expression in kidney biopsies predicts renal function and is a future component of donor scoring criteria

    CDKN2A is a proven and validated biomarker of ageing which acts as an off switch for cell proliferation. We have demonstrated previously that CDKN2A is the most robust and the strongest pre-transplant predictor of post- transplant serum creatinine when compared to “Gold Standard” clinical factors, such as cold ischaemic time and donor chronological age. This report shows that CDKN2A is better than telomere length, the most celebrated biomarker of ageing, as a predictor of post-transplant renal function. It also shows that CDKN2A is as strong a determinant of post-transplant organ function when compared to extended criteria (ECD) kidneys. A multivariate analysis model was able to predict up to 27.1% of eGFR at one year post-transplant (p = 0.008). Significantly, CDKN2A was also able to strongly predict delayed graft function. A pre-transplant donor risk classification system based on CDKN2A and ECD criteria is shown to be feasible and commendable for implementation in the near future

    Highly syntenic and yet divergent: a tale of two Theilerias

    The published genomic sequences of the two major host-transforming Theileria species of cattle represent a rich resource of information that has allowed novel bioinformatic and experimental studies into these important apicomplexan parasites. Since their publication in 2005, the genomes of T. annulata and T. parva have been utilised for a diverse range of applications, ranging from candidate antigen discovery to the identification of genetic markers for population analysis. This has led to advancements in the quest for a sub-unit vaccine, while providing a greater understanding of variation among parasite populations in the field. The unique ability of these Theileria species to induce host cell transformation is the subject of considerable scientific interest and the availability of full genomic sequences has provided new insights into this area of research. This article reviews the data underlying published comparative analyses, focussing on the general features of gene expression, the major Tpr/Tar multi-copy gene family and a re-examination of the predicted macroschizont secretome. Codon usage between the Theileria species is reviewed in detail, as this underpins ongoing comparative studies investigating selection at the intra- and inter-species level. The TashAT/TpshAT family of genes, conserved between T. annulata and T. parva, encodes products targeted to the host nucleus and has been implicated in contributing to the transformed bovine phenotype. Species-specific expansion and diversification at this critical locus is discussed with reference to the availability, in the near future, of genomic datasets which are based on non-transforming Theileria species

    Frogs (Coqui Frogs, Greenhouse Frogs, Cuban Tree Frogs, and Cane Toads)

    Amphibians are perhaps most well known for their highly threatened status, which often masks appreciation for the great numbers of species that are widespread global invaders (Kraus 2009). Both purposeful and accidental introductions of amphibians have occurred worldwide. Motivations for purposeful amphibian introductions include their use as biocontrol agents and culinary ambitions (Storer 1925; Kraus 2009). However, there are an increasing number of amphibians that are being accidentally introduced and becoming widespread (Kraus 2009). These introductions are in some ways more disconcerting because they may be the most difficult to prevent in the future. There are 19 nonnative amphibians that have become successfully established in 28 of the 50 U.S. states (Figure 9.1; Kraus 2009). The most successful non-native amphibian is the bullfrog (Lithobates catesbeianus), which has become established in 19 states outside of its native range on the eastern side of the United States, followed by the Cuban greenhouse frog (Eleutherodactylus planirostris), which has established itself in six states, and five frog species, including the Puerto Rican coqui (E. coqui), which are now established in three states outside of their native range (Figure 9.1; Kraus 2009). The state with the most nonnative frogs is California with eight species, followed by Hawaii with six, and Florida and Arizona with four (Table 9.1; Kraus 2009). Many nonnative amphibians in the United States, particularly in the western United States, are from other parts of the United States, namely, east of the Mississippi River. However, there are also many nonnative amphibians with tropical or subtropical origins that are primarily successful in tropical and subtropical states, such as Florida and Hawaii, and territories, such as Guam

    Managing Roof Rats in Citrus Orchards: Initial Efforts toward Building an Integrated Pest Management Program

    Roof rats cause extensive damage in orchards throughout the world. Integrated Pest Management (IPM) systems are the best option for managing rodents, yet few management systems have been developed and tested to control roof rats in agricultural settings. We initiated a study in 2020 to provide the foundation for an IPM program to manage roof rats in California citrus orchards. Our initial efforts centered on developing effective monitoring strategies for roof rats to determine when management actions are needed, assessing rat movement patterns to determine proper placement of management tools, and conducting initial tests of Goodnature A24 self-resetting traps and elevated bait stations containing 0.005% diphacinone-treated oats. We determined that the use of both tracking tunnels and remote-triggered cameras served as effective monitoring tools for roof rats in citrus orchards, and a smaller 3 × 3 grid placement of these monitoring tools was as effective as a 5 × 5 grid, indicating that substantial material and labor costs could be saved by using the smaller grid size. Placement of the monitoring tools on the ground or up in trees did not influence the effectiveness of this approach. We determined that roof rats exclusively used orchard habitats rather than surrounding fields, so control efforts should be focused in these areas. Roof rats moved substantial distances daily (~170-190 m), but results from bait station trials suggest that spacing of bait stations and traps should be closer together to increase roof rat encounter rates of these devices. Our trial with A24 traps elevated in orchard canopies suggest that there is an advantage to including a platform underneath the trap to increase trap activation. Collectively, this information provides a baseline for future research targeted at developing an effective IPM program for managing roof rats in citrus orchards
